Title: "Centrosomal Localization of TDP-43: New Perspectives on ALS Neurodegeneration"

Abstract: Amyotrophic Lateral Sclerosis (ALS) is an adult-onset neurodegenerative disease affecting motor neurons in the brain and spinal cord. The disease leads to progressive and widespread paralysis involving the limbs, bulbar region, and respiratory muscles. Conformational changes and pathological accumulation of the TDP-43 protein within the nervous system is a hallmark of the disease identified in 2006, which has driven major advances in research over the past two decades. However, our still incomplete understanding of the pathophysiological mechanisms underlying ALS, the wide clinical heterogeneity among patients, and the lack of experimental models that accurately replicate the disease explain the lack of treatments. In this lecture, we will discuss current hypotheses regarding neurodegeneration in ALS, the results of collaborative work with Dr. Anne-Marie Tassin’s team identifying a centrosomal localization of TDP-43 in cells, and the current challenges and future perspectives in ALS research.
