 gers\nAbstract: Amyotrophic Lateral Sclerosis (ALS) is an adult-onset neur
 odegenerative disease affecting motor neurons in the brain and spinal cord
 . The disease leads to progressive and widespread paralysis involving the 
 limbs\, bulbar region\, and respiratory muscles. Conformational changes an
 d pathological accumulation of the TDP-43 protein within the nervous syste
 m is a hallmark of the disease identified in 2006\, which has driven major
  advances in research over the past two decades. However\, our still incom
 plete understanding of the pathophysiological mechanisms underlying ALS\, 
 the wide clinical heterogeneity among patients\, and the lack of experimen
 tal models that accurately replicate the disease explain the lack of treat
 ments. In this lecture\, we will discuss current hypotheses regarding neur
 odegeneration in ALS\, the results of collaborative work with Dr. Anne-Mar
 ie Tassin’s team identifying a centrosomal localization of TDP-43 in cel
 ls\, and the current challenges and future perspectives in ALS research.\n
