Title: Multi-omics dissection of allele-specific regulatory mechanisms at the imprinted Dlk1-Dio3 domain

 

Abstract: Genomic imprinting is an epigenetic mechanism that controls the parent-specific, monoallelic expression of ~150–200 genes in mammals, organized in 20 clusters. This regulation is primarily mediated by parent-specific DNA methylation at CpG-rich regions called Differentially Methylated Regions (DMRs). DNA methylation status at DMRs orchestrates a cascade of allelic regulatory mechanisms, which can vary between imprinted domains, including histone modifications, CTCF-mediated 3D genome architecture and long non-coding RNAs (lncRNAs). In my thesis project, I focused on two mostly unexplored layers; differential replication timing and nuclear lamina association.
